Non-carcinogenic alternative of Ethidium Bromide, DNA Stain can be used as a safer alternative compared to the traditional ethidium bromide stain used for DNA and RNA detection in agarose gel. Compared to ethidium bromide, it has low toxicity and mutagenicity. DNA Stain has two secondary fluorescence excitation peaks (~270 nm; ~295 nm) and one strong excitation peak centered around 490 nm. The fluorescence emission is similar to Ethidium Bromide when bound to DNA which is at about 530 nm.

Usage Protocols

Precasting method:

• • Prepare 100ml of 0.8-3.0% agarose gel solution.
  • Heat till solution is completely clear.
  • Add 4-6µl of DNA stain and mix gently with gel.
  • Cool gel to 50-60°C and cast gel into gel tray.
  • Load samples into solidified gel and perform electrophoresis.
  • Detect bands under UV illuminator.

Post-staining method:

• • For agarose gel less than 0.5cm thickness, 10 – 25µl of the stain should be used per 100ml of buffer
  • Staining time is optimized between 5 – 60 minutes; amount of stain varies with gel thickness and concentration of agarose.
  • Visualize DNA by staining with DNA stain (C/No: BIO-5170-1ml).

Used for loading DNA markers and samples on agarose or polyacrylamide gels. It contains three dyes – Orange G, Xylene cyanol FF and Bromophenol blue. This allows better visual tracking of DNA migration during electrophoresis.
