How do I place a Whole Plasmid Sequencing order? Do I need a quotation before placing an order?
You can place your Whole Plasmid Sequencing order directly through AxillentHub. A quotation is not required to proceed with your order.
What is the expected turnaround time?
Results are delivered within 24 hours of receiving a suitable sample.
How much sequencing coverage will I receive per plasmid?
We are unable to guarantee a specific level of sequencing coverage, as the number of raw reads generated may vary depending on factors such as plasmid length and sample quality. Generally, each plasmid is expected to achieve a minimum coverage of 30×.
How accurate is the long-read sequencing data?
Current Oxford Nanopore sequencing chemistry can achieve raw-read accuracies of up to approximately 99%. Multiple sequencing reads are assembled and polished to generate a highly accurate plasmid consensus sequence.
Will the submitted plasmid undergo QC before sequencing?
No. To maintain a fast turnaround time and competitive pricing, we do not perform QC on plasmid samples prior to sequencing. Customers are advised to ensure that submitted samples meet the specified sample requirements.
Can I submit a bacterial broth or colony instead of purified plasmid DNA?
Currently, Whole Plasmid Sequencing requires purified plasmid DNA. We are exploring options to offer bacterial broth and colony-based submissions in the future.
Is PlasmidLab free to use?
Yes. PlasmidLab is provided free of charge to Whole Plasmid Sequencing users.
What happens if my plasmid fails to assemble? Is a free rerun provided?
Plasmid assembly failure is typically associated with insufficient sequencing reads for the bioinformatics pipeline to generate a consensus sequence. Common contributing factors include:
- Degraded or fragmented DNA
- Insufficient dsDNA concentration
- Mixed plasmid populations
For optimal sequencing results, please follow the Whole Plasmid Sequencing Sample Requirements before submitting your sample.
Free reruns are considered on a case-by-case basis. If you have questions or concerns regarding your sequencing results, please contact ngs@axilscientific.com.
How long will my samples be retained, and will they be returned to me?
Samples will be retained for a maximum of 2 weeks after the sequencing results are released. Any remaining sample after this period will be disposed of and will not be returned to the customer.
