Sample Preparation for Species Barcoding

Please refer below for the sample preparation procedures for Animal Samples, Plant Samples, Bacterial and Fungal Samples.

Sample Preparation of Animal Samples

  1. Slice the tissue sample with a new scalpel blade on a sterile surface (sterile Petri dish).
    Change to new blades for every new sample to be sliced.
  2. Weigh the tissue sample and ensure it is within a range of 10-100 mg.
  3. Transfer 25 µl of Solution TP [Tube TP] to 100 µl of Solution A [Tube A]. Mix well by pipetting.
  4. Transfer the tissue sample to the tube containing 125 µl of Solution A/ Solution TP. Vortex mix the Solution containing the sample briefly (15 to 20 seconds). Make sure that the tissue sample is covered by the Solution A/ Solution TP.
  5. Incubate at room temperature for 10 minutes.
  6. Incubate at 95 o C for 3 minutes in a heat block or water bath.
    Do not exceed 3 minutes. The Incubation time is very critical.  
  7. Add 100 µl of Solution B [Tube B] to the sample in Tube A after heat treatment. Vortex mix to neutralise the reaction. 
  8. Ship the processed sample to us as soon as possible. 
    Store the processed sample at 4 o C if it is to be shipped later. Recommended to ship within a day of processing the samples

 

Sample Preparation of Plant Genomic DNA

  1. Slice 5 mm square of leaf sample with a new scalpel blade. 
    Change to new blades for every new sample to be sliced
  2. Transfer the 5 mm square of leaf sample to the tube containing 100 µl of Solution A [Tube A]. Vortex mix briefly. 
    Make sure that the leaf sample is covered by Solution A.
  3. Incubate at 95 o C for 10 minutes in a heat block or water bath. 
    Do not Exceed 10 minutes since the incubation time is very critical.
  4. Add 100 µl of Solution B [Tube B] to the sample in Tube A after heat treatment. Vortex mix to neutralise the reaction.
  5. Ship the processed sample to us as soon as possible. 
    Store the processed sample at 4 o C if it is to be shipped later. In any case, ship it within a day of processing the samples.

 

Sample Preparation of Bacterial Sample

  1. Prepare your bacterial sample in either way listed as below.
                     - Cut agar (5mm square) contains bacteria in 1.5mL centrifuge tube. Triplicates.
                      

 

                     - Spin down 1 - 2mL of liquid culture in 1.5mL centrifuge tube. The size of the cell pellet is about 50uL. Triplicates.
                      

 

                     - If the sample is pure isolate, please inoculate > 20 colonies in 125 ul sterile water/ buffer (such as TE, PBS).
                      Apply less than 125uL of overnight liquid culture directly on FTA card (chargable). Leave it dry.
                      Then place card in an envelope/plastic bag for sample submission.
